Andrographolide is the main diterpenoid lactone contained in the leaves of Andrographis paniculata. This bioactive component has multifunctional medicinal properties such as activity against fever, dysentery, diarrhoea, inflammation, cancer and sore throat as well as immune disorder. No solvent is ideal for the extraction of Andrographolide, however we can consider 60% methanol for the model development. Cancer is the largest single cause of death in both men and women, claiming over 6 million each year in the world. CML accounts for about 20% cases of adult and childhood leukemia. Andrographolide acts by targeting the tyrosine kinase activity of BCR-ABL which represents a very promising therapeutic strategy in chronic myeloid leukemia (CMML). In the present study, we focused on amplifying the ABL kinase domain sequence and b2a2 junction domain sequence and tried to find out the sequence difference between normal K562 cells and AR-K562 cells .for this we developed resistant cell lines against Andrographolide by gradual increase in the in the concentration of Andrographolide (1nm to 1um), the morphological features were studied in phase contrast microscopy.
