Zaltoprofen is a NSAID class of drug which has excellent effect on post-surgery or post trauma chronic inflammation of the drug. An attempt has been made to prepare the controlled release CPOP tablet for 12hr release kinetics twice a day. Zaltoprofen has low solubility so, an inclusion complex was made by kneading method using HP-?-CD in order to increase solubility of the drug. CPOP tablet containing Zaltoprofen were prepared by direct compression method by using various osmotic agent like sodium bicarbonate, sodium chloride, mannitol and potassium carbonate. Cellulose acetate, Sorbitol and PEG-400 were selected for coating materials. Formulated tablet also passes the various tablet evaluation parameters. Results of in-vitro drug release had indicated that as the amount of osmotic agent increased, amount of drug release increased and increased in %weight gain decreased the %drug release. Batch Z12 has shown 98.08% drug release compare to other batches so, accepted as optimized batch and evaluated by different pharmacokinetic models. The results of these models have shown that the batch Z12 follows zero order release kinetics and independent of the pH and agitational intensity.
